== Applications == Despite the reduced efficiency verses reversed phase HPLC, hundreds of applications have been reported using MLC. One of the most advantageous is the ability to directly inject physiological fluids. Micelles have an ability to solubilize proteins which enables MLC to be useful in analyzing untreated biological fluids such as plasma, serum, and urine. Martinez et al. found MLC to be highly useful in analyzing a class of drugs called b-antagonists, so called beta-blockers, in urine samples. The main advantage of the use of MLC with this type of sample, is the great time savings in sample preparation. Alternative methods of analysis including reversed phase HPLC require lengthy extraction and sample work up procedures before analysis can begin. With MLC, direct injection is often possible, with retention times of less than 15 minutes for the separation of up to nine b-antagonists. Another application compared reversed phase HPLC with MLC for the analysis of desferrioxamine in serum. Desferrioxamine (DFO) is a commonly used drug for removal of excess iron in patients with chronic and acute levels. The analysis of DFO along with its chelated complexes, Fe(III) DFO and Al(III) DFO has proven to be difficult at best in previous attempts. This study found that direct injection of the serum was possible for MLC, verses an ultrafiltration step necessary in HPLC. This analysis proved to have difficulties with the separation of the chelated DFO compounds and with the sensitivity levels for DFO itself when MLC was applied.
The second phase uses a modified MaxSub algorithm: a single 7 reside aligned pair in each proteins is used to orient the two full length protein structures to maximally superimpose these just these 7 C-alpha, then in this orientation it scans for any additional aligned pairs that are close in 3D. It re-orients the structures to superimpose this expanded set and iterates until no more pairs coincide in 3D. This process is restarted for every 7 residue window in the seed alignment. The output is the maximal number of atoms found from any of these initial seeds. This statistic is converted to a calibrated E-value for the similarity of the proteins. Mammoth makes no attempt to re-iterate the initial alignment or extend the high quality sub-subset. Therefore, the seed alignment it displays can't be fairly compared to DALI or TM align as it was formed simply as a heuristic to prune the search space. (It can be used if one wants an alignment based solely on local structure-motif similarity agnostic of long range rigid body atomic alignment.) Because of that same parsimony, it is well over ten times faster than DALI, CE and TM-align. It is often used in conjunction with these slower tools to pre-screen large data bases to extract the just the best E-value related structures for more exhaustive superposition or expensive calculations.
=== Neurotransmission === Agmatine has been discussed as a putative neurotransmitter. It is synthesized in the brain, stored in synaptic vesicles, accumulated by uptake, released by membrane depolarization, and inactivated by agmatinase. Agmatine binds to α2-adrenergic receptor and imidazoline receptor binding sites, and blocks NMDA receptors and other cation ligand-gated channels. However, while agmatine binds to α2-adrenergic receptors, it exerts neither an agonistic nor antagonistic effect on these receptors, lacking any intrinsic activity. Apart from an absence of known agmatine-specific post-synaptic receptors, agmatine fulfills Henry Dale's criteria for a neurotransmitter and is hence considered a neuromodulator and co-transmitter. The existence of theoretical agmatinergic-mediated neuronal systems has not yet been demonstrated although the existence of such receptors is implied by its prominence in the mediation of both the central and peripheral nervous systems. Research into agmatine-specific receptors and transmission pathways continues. Due to its ability to pass through open cationic channels, agmatine has also been used as a surrogate metric of integrated ionic flux into neural tissue upon stimulation. When neural tissue is incubated in agmatine and an external stimulus is applied, only cells with open channels will be filled with agmatine, allowing identification of which cells are sensitive to that stimuli and the degree to which they opened their cationic channels during the stimulation period.

== History == Avasimibe was the result of a rational drug design process carried out at Parke-Davis in the early 1990s which sought to obtain orally bioavailable, water-soluble ACAT inhibitors; all such inhibitors known at the time were lipophilic and poorly absorbed when taken by mouth. This process yielded several compounds with potential, including one (designated PD 138142–15) with good solubility in water and remarkable efficacy in animal studies, but it was chemically unstable and degraded rapidly, especially in acidic environments. (Undesirable CYP450 induction was first noted at this time, in PD 138142-15 and its degradation products.) Chemical modification of PD 138142-15 and retrosynthetic analysis found that avasimibe (then codenamed CI-1011) could be easily manufactured from commercially available starting compounds, and once its efficacy was demonstrated in vitro and in rat studies, it was selected for further development. After additional safety and preclinical efficacy studies in animals, phase I clinical trials in humans began in 1997, first for hyperlipidemia (June) and subsequently for atherosclerosis (December). Phase II trials for both indications followed in 1998, and phase III trials in 2001. In October 2003, clinical development of avasimibe was discontinued. Later research discredited the concept of ACAT inhibition as a treatment for dyslipidemia and atherosclerosis, and interest in these compounds as a class waned accordingly.
